Human cRAF
Human cRAF Xpress Clones express cRAF (Raf-1), a serine/threonine kinase regulated by RAS GTPases and involved in mediating cellular responses to growth signals. cRAF activates MEK1/MEK2, which in turn phosphorylate ERK1/ERK2, key players in cell physiology, influencing gene expression, cell division, apoptosis, differentiation, and migration. The c-terminal kinase domain of cRAF (residues 305-648) is cloned into the pMEV-2HA vector. The K375M mutation, which alters the catalytically essential lysine to methionine, renders the cRAF kinase inactive. This kinase-dead mutant can inhibit endogenous cRAF activity, serving as a dominant-negative inhibitor in mammalian cells.
