Human cRAF Xpress Clones express cRAF (Raf-1), a serine/threonine kinase that plays a pivotal role in RAS signaling. cRAF activates MEK1/MEK2, which then phosphorylate and activate ERK1/ERK2, critical effectors in regulating cell cycle, apoptosis, differentiation, and migration. The c-terminal kinase domain of cRAF (residues 305-648) is cloned into the pMEV-2HA vector. The K375M mutation, which alters the essential lysine to methionine, renders cRAF kinase-inactive and allows it to act as a dominant-negative inhibitor of endogenous cRAF activity in transfected mammalian cells.
